Default p0301 code on a newly rebuilt motor in 01 xj

well i got that famous misfire code..i have literally 35 miles on this motor....i replaced the ecm because the ground trigger was gone on #1 injector...now i cant get the p0301 code off....ive swapped the injectors to cylinder #2 no change ....i also believe i didnt get a lifter in #1 cylinder completly pumped up...could that be my issue......how could i pump the lifter up without removing the head????
list the possible solutions for this please that way i can trouble shoot it starting with the cheapest

For a single cylinder misfire, I always approach it in this manner....

1. Spark plug. Swap it out, even if it's new.
2. Spark plug wire. Swap it.
3. Distributor cap. Change it, even if it's new
4. Injector (you already swapped that)
5. Internal condition of that cylinder. Start with a compression test and see how that cylinder reads in comparison with the others.

Also, did you rebuild it? Did you install a remanufactured? If there is warranty, don't be afraid to use it!

i completly rebuilt it my self ..94 block. bought a re ring kit that included rings rod an mains, all gaskets , oil pump, timing set & guide, camshaft along with camshaft bearings, lifters an pushrods, .. head an valves are orginal along wit the springs....an this doesnt have a distributor so that rules that out.... ive tired to champion coppers on #1 cyl. an it doesnt effect it

an could the camshaft sensor be out of sync cause this issue..?? it has not been on a scan tool since the rebuild
