if by 4 down you mean all wheels on the road, then yes.
it's easier with a manual tranny, but you should still put the t-case in neutral.
with 4wd, all wheels should be up or down.

some jeep models it is not advisable to pull the rear shaft due to the slip yoke, the d shaft is what seals the fluid in the t case, with no shaft, the fluid can come out of the t case.

2wd and a standard tranny? no problem then. just put it in neutral and you should be good to go.
it's the autos that need to be kept lubed when the wheels are turning and the engine is off. so that's why the t-case needs to be in neutral and the tranny in park. that way nothing is turning in the tranny to dry out the torque converter seal and cause internal leaking. happened to me once. not fun changing it out in a campground on the grass.
